On Monday, Messi's Ligue 1 champions announced that he had not recovered from a calf injury.
The 35-year-old was unable to participate in the league's draw with Stade de Reims last weekend due to an injury.
Galtier told reporters on Monday, "We know the importance of Leo in our game, his form and the connection he can have with other players.
"His absence can't make us happy. However, we will need to make additional connections in order to pose a threat to the Benfica defense.
“We must have other options." We need to be able to play on both sides. When Messi plays, we play a lot down the middle. We’ve been working hard on distribution to add more variety to our attack.
